内容記述 | Alcohol drinking is supposed to be one of risk factors of atherosclerosis. The risk of alcohol intake was mainly evaluated in male subjects, because of high percentage of drinking population in male. However, atherosclerosis is a typical disease that has sexual difference in morbidity and mortality. Therefore, we investigated whether ethanol causes different effects on macrophages, which play a pivotal role in the initiation and the development of atherosclerosis, from different sex origin. Peritoneal macrophages were isolated from rats stimulated with lipopolysaccharide. NO production was measured as an index of macrophage activation by Griess reagent method (NO_3^-/NO_2^- concentration) and NO sensitive fluorescent dye (relative NO concentration). Ethanol reduced NO_3^-/NO_2^-concentration from500mM and NO production from 300mM in macrophages from male rats. Ethanol reduced both parameters in macrophages from female rats in less extent. The addition of 17β-estradiol to macrophages suppressed the reduction of NO production from macrophages by ethanol. However, there was no statistical difference in both experiments. These results suggest that ethanol reduces the activation of macrophages and may suppress the progression of atherosclerosis. Although the protective effect of ethanol was limited in female rats compared with male rats, there was no significant sexual difference in our model. | |||||
